Optogenetic modulation of serotonin release in the dorsal hippocampus
- The serotonergic system is closely associated with the modulation of the affective state and is known to influence memory acquisition within the hippocampus. The aim of this study was to analyze how serotonin (5 HT) can change information processing within the hippocampus. Behavioral tests were performed in combination with optogenetic manipulation of serotonergic release in the mouse hippocampus. Serotonergic release within the dorsal cornu ammonis 1 (CA1) was optogenetically stimulated via Channelrhodopsin 2 (ChR2) or reduced via the exogenous 5 HT\(_{1B}\) receptor chimera (vSWO 5 HT\(_{1B}\)). A second experimental approach investigated the specific activation of an exogenous 5-HT\(_{1A}\) receptor chimera (vLWO-5-HT\(_{1A}\)) on pyramidal neurons. Finally, the results show that only spatial memory, and not object recognition, is affected by the modulation of serotonergic signaling, indicating a key role of serotonin in processing spatial information in the hippocampus.
- Das serotonerge System ist eng mit der Modulation des affektiven Zustands verbunden und beeinflusst nachweislich den Gedächtniserwerb im Hippocampus. Ziel dieser Studie war es zu analysieren, wie Serotonin (5-HT) die Informationsverarbeitung im Hippocampus verändert. Es wurden Verhaltenstests in Kombination mit der optogenetischen Manipulation der Serotoninfreisetzung im Hippocampus der Maus durchgeführt. Die Serotoninausschüttung in serotonergen Axonen in CA1 wurden entweder mit Hilfe von Channelrhodopsin 2 optogenetisch stimuliert oder mit Hilfe der 5-HT\(_{1B}\)-Rezeptorchimäre (vSWO 5 5-HT\(_{1B}\)) gehemmt. In einem zweiten Experiment wurde die Aktivierung der exogenen 5-HT\(_{1A}\) Rezeptor-Chimäre (vLWO- 5-HT\(_{1A}\)) auf Pyramidenzellen untersucht. Schließlich zeigten die Ergebnisse, dass nur das räumliche Gedächtnis und nicht die Objekterkennung durch die optogenetische Stimulation beeinflusst wurden, was auf eine Schlüsselrolle von Serotonin bei der Verarbeitung räumlicher Informationen hinweist.
