Investigation of the effect of novel spatial information processing on hippocampal neuronal networks
- The role of the hippocampus in learning and memory is well described. It is unknown, to what extent the subfields of this brain structure contribute to neuronal activity during learning. Here, nuclear expression of immediate-early gene was mapped in the dorsal hippocampus subfields. This reflects the neuronal population and circuits that are recruited during neural activity. Whereas a weak afferent stimulation induces no changes in the nuclear expression of \(\it H1a\), a prolonged afferent stimulation significantly increases \(\it H1a\) expression across the dorsal hippocampal subfields. Exposure to a novel empty space activates all subcompartments in the dorsal hippocampus. Interestingly, acquisition of novel spatial content triggers a subcompartment-specific expression of \(\it H1a\). In the animal model of Alzheimer’s Disease, the ability to acquire novel memory is still evident, however, significantly impaired when compared to healthy condition.
- Die Rolle des Hippocampus für das Lernen und die Gedächtnisbildung ist hinreichend bekannt. Die Beteiligung der Subregionen an neuronalen Aktivitäten während des Lernvorgangs wurde bisher nicht beschieben. Hier wurde die intranukleäre Expression der immediate-early genes in den Subregionen des dorsalen Hippocampus aufgezeichnet. Diese Expression entspricht der neuronalen Population und Netzwerke, die während der neuronalen Aktivität rekrutiert werden. Eine anhaltend, aber nicht schwache, afferente Stimulation führt zu einer signifikanten Erhöhung der \(\it H1a\) Expression. Exploration eines neuen leeren Raums aktiviert alle Subregionen. Interessanterweise führt die Erkundung des neuen Rauminhalts zu einer Subcompartment-spezifischen Expression von \(\it H1a\). In einem Alzheimer-Tiermodell sind die Ratten durchaus noch in der Lage, neue Information aufzunehmen, auch wenn sich diese Fähigkeit im Vergleich zu gesunden Tieren signifikant verschlechtert hat.
