Signalling events evoked by hypertonic stress
- Monolayers of primary rat hepatocytes served as an epithelial model system for the investigation of hypertonicity-induced signalling events towards the restoration of cell volume and shape. Electrophysiological techniques were used to monitor the activation of \(Na^{+}\) conductance, the main mechanism of regulatory volume increase following hypertonic cell shrinkage, in combination with a pharmacological approach. This study provided further evidence in support of the hypothesis that exocytosis of \(Na^{+}\) channels mediates the activation of \(Na^{+}\) conductance. Furthermore, it could be shown that PLC, but not PI-3 kinase is essential in orchestrating \(Na^{+}\) conductance increase. With biochemical approaches, focal adhesion signalling could be shown to be affected by hypertonic stress. Furthermore, a shift in the activities of the small GTPases Rac and Cdc 42 towards Rho A was observed. In addition, hypertonicity induced two phases of F-actin formation, i.e. a Rho kinase-dependent and -independent phase.
- An primären Ratten-Hepatozyten wurden hyperton-induzierte Signalereignisse der Regulation von Zellvolumen und -form untersucht. Es konnten hinweise darauf erhalten werden, dass die Aktivierung der \(Na^{+}\) Leitfähigkeit, dem Hauptmechanismus der Volumenregulation nach hypertoner Schrumpfung, durch Exozytose erfolgt. Zudem konnte gezeigt werden, dass die PLC, nicht aber die PI-3 kinase, Bestandteil der Aktivierungskaskade ist. Hypertoner Schock induziert die Aktivierung der Tyrosin Kinasen Src und FAK, Komponenten der Fokaladhäsionen. Zudem kommt es zu einer Verlagerung der Aktivität kleiner GTPasen von Rac und Cdc 42 zu Rho A kommt. Hypertoner Stress induzierte die Bildung von F-Aktin in Rho Kinase abhängiger und unabhängiger Weise. Aufgrund dieser Befunde konnte ein Modell entwickelt werden, welches die Regulation der Zellform nach hypertonem Stress erklärt. Darüber hinaus wurde eine Arbeitshypothese erstellt, welches die Regulationswege von Zellform und Zellvolumen integriert.
