Physiological adaptation of the membrane proteome to aromatic compounds and steroids as alternative carbon sources in \(\it {Corynebacterineae}\)
Physiologische Adaption des Membranproteoms an Aromaten und Steroide als alternative Kohlenstoffquellen in \(\it {Corynebacterineae}\)
- Actinomycetes can use aromatic compounds and steroids as sole carbon/ energy sources. Aromatic compounds are renewable resources with potential biotechnological applications, while cholesterol catabolism is important during \(\textit {Mycobacterium tuberculosis}\) infection. Both compounds can interact with membranes, yet only scarce knowledge exists about their impact on membrane proteins. Quantitative proteomics was used to analyze the membrane proteomes of \(\textit {Corynebacterium glutamicum}\) grown on aromatic compounds and \(\textit {Rhodococcus jostii}\) grown on steroids. In benzoate-grown \(\textit {C. glutamicum}\), 2 co-expressed transporters may both contribute to benzoate uptake. Also, energy and amino acid metabolism were affected. Growth of \(\textit {R. jostii}\) on cholesterol and cholic acid led to expression of different degradation enzymes. Also, 9 subunits of the cholesterol transport system were quantified. Proteomics provided insight into global adaptation processes to alternative carbon sources and compound-specific responses.

Aromaten sind nachwachsende Rohstoffe mit potentiellen Anwendungen in der Biotechnologie; Cholesterin-Abbau in \(\textit {R. jostii}\) dient als Modell für das pathogene Bakterium \(\textit {Mycobacterium tuberculosis}\). Obwohl Aromaten und Steroide mit Membranen interagieren können, ist ihre Auswirkung auf Membranproteine bisher kaum untersucht. Mithilfe quantitativer Proteomik wurde die Adaption der Membranproteome von \(\textit {C. glutamicum}\) an Aromaten und von \(\textit {R. jostii}\) an Steroide untersucht. Benzoat bewirkte in \(\textit {C. glutamicum}\) die Coexpression zweier Benzoat-Transporter, sowie Adaption des Energie- und Aminosäuremetabolismus. Cholesterin und Cholsäure induzierten in \(\textit {R. jostii}\) die Expression verschiedener Abbau-Enzyme. Auch wurden 9 Untereinheiten des Cholesterin-Transporters quantifiziert. Proteomik ermöglichte sowohl die Detektion globaler, als auch Substrat-spezifischer Adaptions-Prozesse.
